基于纸基的荧光碳量子点传感器的可视化检测研究进展

摘要: 碳量子点作为一种新型的纳米材料,其独特的荧光性能使其在物质检测领域得到了越来越多的关注。利用碳量子点与物质反应导致荧光强度发生变化的特性,可将其应用于可视化检测,检测方法分为荧光猝灭型和荧光恢复型。基于纸基的荧光传感器与基于液相反应的荧光检测相比,其快速、现场和可视化的特点更加突出,因此有着更为广泛的应用。本文针对碳量子点的荧光特性、可视化荧光检测、纸基传感器的研究现状,并结合本课题组相关研究进展进行论述,以期为今后纸基碳量子点荧光传感器的研究和应用提供参考。

Research Progress of Paper-based Fluorescent Sensor on Carbon Quantum Dots for Visual Detection

Abstract: Carbon quantum dots (CQDs), as a new class of nanomeaterials, have exhibited broad appeal for chemical inspections according to their peculiar fluorescent properties. When interacting with detected molecules, their tunable fluorescence intensity can be applied in visual detection which includes FQ-type (fluorescent quenching) and FR-type (fluorescent recovery) detection. Compared with aqueous fluorescent sensors, paper-based devices are more extensively utilized in the field of substance inspections concerning their superiorities in portability, fast-response, on-site examination and visualization. Herein, we review recent research status and progress in the field of CQDs to provide a future perspective incorporating fluorescent properties of CQDs, current visual fluorescent inspections and paper-based sensors.

碳量子点(Carbon quantum dots,CQDs)是一种粒径小于10 nm,具有一定分散性的类球形碳纳米颗粒,于2004年Xu等在电泳法制备单壁碳纳米管的纯化过程中首次被发现[1];2006年,Sun等[2]通过表面钝化合成了具有增强荧光发射效应的荧光纳米颗粒,并将其命名为碳量子点。与传统半导体量子点相比,CQDs不仅具有类似传统量子点的发光性能与纳米尺寸特性,而且具有较好的水溶性、化学惰性、低毒性、高耐光漂白、易功能化和良好的生物相容性等特点,因而在不同领域获得了广泛关注与应用[3]

CQDs的光学性能是其最受关注的特性,它在紫外光区及可见光区有宽且连续的光吸收,并且具有较大的激发光波长范围。此外,CQDs表面具有大量的活性基团,可与多种物质发生螯合反应,使其荧光强度发生改变,利用这一性能,可将CQDs应用于荧光检测领域[4]。用CQDs制成的荧光传感器具有荧光强度高、荧光稳定性强、可视化好,检测方便等优点,可应用于多种物质的检测分析(图 1)[5]

与液相荧光检测相比,基于纸基的荧光传感器的便携、快速、现场和可视化的特点更加突出,因而在物质检测中被广泛应用。基于此,许多研究工作者根据待检测物质的性质制备了对其进行专一检测的荧光检测试纸[6-10]。本文将介绍CQDs的荧光特性及其近年来在物质检测中的应用,此外,对在纸基上实现CQDs可视化检测的研究进展,以及本课题组的相关研究成果进行介绍。

1 碳量子点的荧光特性 1.1 碳量子点的发光性质

典型的CQDs在紫外光区(230~320 nm)有很强的吸收带,且吸收谱带一直延伸至可见光区[11],同时也伴随一些小肩峰的产生,这一现象可归因于C=C键的π-π*跃迁或C=O键的n-π*跃迁。通常表现出荧光最大发射波长、强度依赖激发波长等光学特征[12, 13]。如图 2所示,当激发波长从290 nm增长到450 nm时,CQDs的最大发射峰由415 nm变化至540 nm,充分表明CQDs具有一定的激发波长依赖性[14]。值得注意的是,CQDs的表面基团可以轻微调节其吸收带的位置,改变其荧光强度和颜色,并赋予其分析检测物质的能力[3]

CQDs的荧光稳定性决定其在化学传感领域中的应用潜力,这通常包括抗光漂白性和储存稳定性[15]。绝大部分有机荧光染料随光照时间的增加,其荧光强度会逐渐减弱,而CQDs则不然。Zhao等研究表明,CQDs在激光持续照射数小时后,其荧光强度基本不发生变化,也不会产生光漂白和光眨眼等现象[16, 17]。Tao等[18]发现CQDs经过350 W的高亮冷光持续照射24 h后仍可表现出很好的稳定性,而相同实验条件下的荧光素在照射6 h时荧光则几乎完全消失。

1.2 荧光机理与影响因素

已报道的CQDs的发光机理有不同尺寸纳米粒子的量子效应[19]、缺陷和表面态[20]、表面基团[21]等。Zhao等[16]认为,CQDs表面发射能量阱的量子限域效应是其荧光产生的主要原因,且高荧光量子产率通常由小尺寸的CQDs获得,与CQDs表面缺陷无关。相反,Bao等[22]根据CQDs不依赖于尺寸的吸收峰红移现象和表面氧化相关的荧光性质,推断出CQDs的荧光是由其表面缺陷导致。而Hu等[23]认为是由于CQDs纳米粒子表面存在羧基,在激发光照射下,产生含碳、含氧的自由基引起的。由此可见,由于CQDs的结构复杂多样,有关发光机理的研究还未形成完整体系。

影响CQDs荧光强度的因素有很多,首先溶液的pH值对CQDs的荧光强度影响较大,且不同CQDs受pH的影响程度也有较大差异。Zhao等[16]发现溶液中pH值的轻微改变都会引起CQDs荧光强度的改变。而Liu等[24]发现溶液的pH从5降到9时,CQDs的量子产率仅仅下降了大约3%。由于钝化能使CQDs表面的能量带隙得到稳定,让其更容易被激发,因而CQDs在钝化后发光强度会增强[2]。此外,CQDs的尺寸、表面性质、官能团和温度等都是影响荧光性能的重要因素[525]

2 基于碳量子点的荧光可视化检测 2.1 碳量子点的荧光猝灭型可视化检测

CQDs的荧光猝灭型可视化检测方法主要机制有自旋轨道耦合、能量转移和电子转移等[26-28],该方法简单易行,但是灵敏度和选择性较低,在实际检测中易受环境因素的干扰,常应用于Hg2+[29-32]、Fe3+[33, 34]、Cu2+[35-43]、Cd2+[44, 45]等无机金属离子和日落黄[46]、青霉素[47]等有机物的定量检测。

利用CQDs可视化检测金属离子的首次应用是对溶液和活细胞中的Hg2+的检测。由于Hg2+等重金属离子对环境和人体健康都有着严重的危害,因此对其检测方法的探寻是近年来的研究热点。Goncalves等首次证明CQDs溶液和CQDs溶胶-凝胶的荧光强度对Hg2+的存在都有着灵敏的感应[29, 30]。Gao研究组使用N掺杂的CQDs(nitrogen-doped carbon quantum dots,NCQDs)选择性灵敏地检测水相中的Hg2+,并证实了Hg2+可使NCQDs表面状态发生变化,检测限为0.23 μmol/L,线性范围为0~25μmol/L[31]。Mohapatra等[32]设计了基于氮硫共掺碳点(nitrogen and sulphur co-doped carbon dots,NSCDs)的高选择性光致发光探针用于Hg2+的检测,该荧光探针对Hg2+具有很好的特异性和选择性,检测限为0.05 nmol/L,将其应用于HaCaT细胞中Hg2+的检测,具有明显的效果(图 3)。

铁离子(Fe3+)作为环境和生物系统中无处不在的金属离子,也可通过CQDs的荧光猝灭达到灵敏的检测效果。Yu等[33]通过简单且低成本的一锅法合成氮掺杂碳量子点(NCQDs)作为荧光探针灵敏和选择性地检测Fe3+,并证实了Fe3+与NCQDs间电子转移的动力学猝灭机制,其检测限为4.67 μmol/L。Xu等[34]首次使用柠檬酸钠和硫代硫酸钠作为前体,水热法合成了具有高荧光量子产率(67%)的硫掺杂CQDs,并将其用作Fe3+探针,检测范围为1~500 μmol/L,检测限为0.1 μmol/L。

近几年,利用CQDs的荧光猝灭检测铜离子(Cu2+)有着广泛的应用,所制CQDs对Cu2+的检测效果总结于表 1

利用CQDs和食品着色剂日落黄(Sunset yellow,SY)之间的荧光共振能量转移(fluorescence resonance energy transfer,FRET),日落黄可以选择性地猝灭CQDs的荧光。Sun等通过在空气中加热N-(2-羟乙基)乙二胺三乙酸来制备荧光CQDs,并设计了浓度范围为0.3~8.0 μmol/L的日落黄的检测方法,检出限为79.6 nmol/L,对饮料样品中的日落黄获得了令人满意的检测效果[44]

2.2 碳量子点的荧光恢复型可视化检测

CQDs的荧光恢复型可视化检测的主要机制有光诱导电子转移[48]、罗丹明结构互变[49]和聚集诱导[50]等,其与荧光猝灭型相比,实现过程较为复杂,但是灵敏度和选择性较高,能达到更低的检出限。鉴于CQDs的低细胞毒性和较好的生物相容性,CQDs荧光恢复型的检测方法常应用于葡萄糖[51]、抗坏血酸[52]、三聚氰胺[53]、谷胱甘肽[54, 55]等物质的分析检测。

抗坏血酸(ascorbic acid,AA)是一种重要的小生物分子,在许多生化过程中起着关键作用。Hu等[52]通过在CQDs溶液中原位合成MnO2纳米片构建纳米复合材料,MnO42 -由于与CQDs的荧光共振能量转移而使其荧光猝灭并形成MnO2纳米片。一旦将AA引入系统,MnO2纳米片被还原为Mn2+使CQDs荧光恢复。该方法对AA具有快速灵敏的响应,检测时间为2 min,检测限为68 nmol/L。

三聚氰胺(melamine)由于含有多氮杂环易与Hg2+发生配位,当三聚氰胺存在时,Hg2+对CQDs的猝灭作用将被减弱。Lei等[53]借此建立了一种三聚氰胺的检测方法,检测限为0.3 μmol/L,远低于美国食品和药物管理局规定的三聚氰胺安全限制值(20 μmol/L)。使用传感系统对包括原料奶和奶粉在内的多个实际样品进行分析,得到了令人满意的回收率。

Shi等[55]建立了一种由AuNPs和荧光CQDs组成的谷胱甘肽(glutathione,GSH)传感系统。将CQDs加入AuNPs胶体溶液中诱导AuNPs和CQDs的聚集,导致AuNPs由红色变为蓝色。而GSH的存在可以保护AuNPs不被聚集并扩大粒子间距离,使AuNPs的颜色变化和荧光强度得到恢复,该纳米传感器表现出对GSH的高度选择性,检测限低至50 nmol/L,可应用于复杂生物环境中对GSH的敏感检测(图 4)。

3 纸基的荧光可视化检测 3.1 纸基荧光传感器

Chen等[6]将荧光氧化石墨烯作为油墨,在聚偏氟乙稀微孔滤膜上打印出一定的英文字样制得F-检测试纸。检测时,先将试纸浸入一定浓度的MPBA-AgNPs溶液(猝灭剂)中,再将其浸入到不同浓度的F-溶液,取出后在365 nm波长的紫外光照射下观察,随着F-浓度的增大,滤膜上文字的蓝色荧光逐渐增强。此方法不需要光谱仪器和复杂的制样过程,可以现场、快速和敏感地检测F-(图 5)。

Zhang等[7]利用发射红色和绿色荧光的两种不同尺寸的CdTe量子点与二氧化硅纳米颗粒以不同方式结合形成双发射荧光混合纳米颗粒。将探针固定在滤纸上,可在紫外灯下通过黄绿色背景的颜色变化检测不同表面上的痕量三硝基甲苯(2, 4, 6-trinitrotoluene,TNT)颗粒。当TNT存在时,多胺官能化的绿色量子点能够选择性结合TNT使绿色荧光猝灭,而红色荧光量子点则不然,因此纸基上的探针将从黄绿色变为红色。该方法显示出高选择性和灵敏度,马尼拉薄膜上的检测限低至5 ng/mm2,并且肉眼可见(图 6)。

Liu等[8]以ZnO纳米粒子为探针,实现纸基Cu2+传感器的定量检测功能。通过将滤纸浸渍在经Cu2+荧光猝灭的ZnO溶胶中获得便携式Cu2+试纸。基于荧光强度与数字信号(灰度级)之间的映射关系用灰度替代荧光强度,将制作的Cu2+试纸的检测功能从肉眼定性检测扩展到定量检测,检测范围为10~1000 μmol/L(图 7)。

3.2 碳量子点纸基荧光传感器

尽管CQDs荧光检测和荧光纸基传感器都已在物质检测方面得到了十分广泛的应用,但是将二者结合,制备以CQDs为荧光物质的纸基传感器实现物质检测,在国内外并不多见。

Tian等[56]发明了一种可视荧光纸传感器,通过将识别分子连接到CQDs的表面,可准确地在现场检测地下水中的2, 4, 6-三硝基甲苯(TNT)。通过CQDs中的伯氨基与TNT炸药之间的光致电子转移效应,荧光被选择性地猝灭。由此将连接聚乙烯醇(polyvinyl alcohol,PVA)的CQDs滴加在普通滤纸上制成纸基传感器。检测时将其浸入到TNT溶液中,在激发波长为365 nm的紫外灯下,可观察到试纸的荧光强度随着TNT的浓度增加而减弱。该发明中的CQDs荧光纸基传感器检测限为0.213 μmol/L,具有很强的现场检测地下水中TNT残留量的能力,并且十分便携高效(图 8)。

Liu等[57]发明了一种新型的双色碳点比率荧光试纸,通过灵敏的颜色变化实现对铜离子(Cu2+)的半定量分析。合成的红色碳点(r-CDs)表面残余的对苯二胺能能有效地结合Cu 2+产生强烈的可见吸收,并与蓝色碳点(b-CDs)的发射重叠;Cu 2+通过与r-CDs和b-CDs的表面配体的双配位相互作用而使b-CDs吸附到r-CDs表面。这两种机制导致特定的光谱能量转移,猝灭b-CDs的荧光。使用r-CDs和b-CDs(1:7)的混合物作为油墨,通过在纸上喷墨打印制成比率荧光试纸,随着Cu2+离子的添加,试纸颜色呈现从蓝色到橙红色的连续变化,检测限低至25 nmol/L(图 9)。

3.3 本课题组相关研究

对CQDs纸基荧光传感器的研究是一个十分新颖的课题,本课题组也对基于纸基的荧光CQDs可视化检测进行了一定的研究:

(1) 对CQDs的合成、特性、改性和应用等方面进行综述,并重点研究了CQDs的上转换发光性质[17]。对目前采用全印刷制备的或纸质的传感器及基于石墨烯量子点的传感器进行综述,并提出了一种基于石墨烯量子点的全印刷纸质生物传感器[58]

(2) 完成了谷胱甘肽的纸基荧光传感器的初步制备。用柠檬酸和组氨酸为碳源制备CQDs原液,与2 mmol/L的Cu2+溶液以V(CQDs):V(Cu2+)=1:3制成混合荧光探针溶液。将蜡笔涂覆于滤纸表面,留出空白区域作为检测区,将其烘干以使蜡更好地固着于滤纸表面。将涂蜡滤纸浸泡于荧光探针溶液中2 h后取出烘干,检测试纸制备完成。

谷胱甘肽(GSH)可使由Cu2+猝灭的CQDs荧光得到恢复,基于此,用该试纸对GSH进行可视化检测:取5 μL不同浓度的GSH溶液滴加在试纸表面,干燥5 min后在365 nm波长的紫外灯下观察试纸的荧光强度变化(图 10)。

本课题组制备的GSH检测试纸检测范围约为500 μmol/L~20 mmol/L,检测限低至50 μmol/L。基于纸基的GSH检测方法实现了对GSH的便携、高效的可视化检测,在食品、医药等领域具有很好的应用前景。此外,该试纸利用蜡的疏水作用有效地克服了待测液体滴加到试纸上将附着于纸上荧光探针排开的问题,增加了纸基传感器的稳定性。本课题组下一步的工作将从以下3个方面展开:(1)改进试纸的结构,从而扩大检测范围,提高检测灵敏度;(2)尝试用喷墨打印的方式制备CQDs荧光试纸;(3)制备疏水CQDs,以扩大检测物质的范围。

4 结语

CQDs具有许多优于半导体量子点的性能,近年在不同研究领域获得了越来越多的关注与应用。制成的荧光传感器荧光强度高、荧光稳定性强、检测方便、响应时间短、离子选择性好。CQDs的荧光检测可分为荧光猝灭型和荧光恢复型,两者由于检测原理不同,可应用于物质检测的不同领域:前者常应用于Hg2+、Fe3+、Cu2+、Cd2+等无机金属离子和日落黄、青霉素等有机物的检测;后者则常应用于葡萄糖、抗坏血酸、三聚氰胺、谷胱甘肽等物质的检测。

基于纸基的荧光检测,与液相检测相比,其快速、现场和可视化的特点突出,因此应用范围更加广泛。尽管CQDs荧光检测和纸基传感器受到人们的重视,但将二者结合实现基于纸基的荧光CQDs可视化检测却还是一个较为新颖的研究课题。本课题组将其作为主要的研究方向,进行了一些相关实验并初步完成了谷胱甘肽纸基传感器的制备,制备的GSH试纸检测范围约为500 μmol/L~20 mmol/L,检测限低至50 μmol/L。该方法有效地增加了纸基传感器的稳定性,实现了对GSH的便携、高效的可视化检测。

为了提高CQDs的荧光纸基传感器的检测范围和灵敏度,还需要在纸基的稳定性、CQDs和待测物溶液浓度的控制、可视化定量检测中荧光强度与待测物浓度的线性拟合等方面进一步深入研究,这也是未来该领域的研究重点和难点。
