Application Research and Sharing Management of Stochastic Optical Reconstruction Super Resolution Microscope

Abstract: Stochastic Optical Reconstruction Microscope (STORM) uses a single molecule localization algorithm based on optically controlled fluorescent molecules to achieve lateral and axial resolutions up to (~20 nm,~50 nm) respectively. Based on use and management experience, this article introduces the principle of the super resolution STORM microscope, software and hardware components and technical parameters, sample preparation method, the common operation process of super resolution image acquisition, environmental requirements, as well as daily maintenance,management of sharing platform and the applications of STORM microscope. The further improvement of STORM spatial and temporal resolution is prospected.
