虎杖甙/HP-β-CD包合物的制备及光稳定性研究

Preparation and Photostability of the Inclusion Complex of HP-β-CD with Piceid

以羟丙基-β-环糊精(HP-β-CD)为主体,采用冷冻干燥法制备了虎杖甙的包合物,通过相溶解度法、粉末X 射线衍射谱图(XRD)、热重分析(TG)、差示扫描量热法(DSC)、扫描电子显微镜(SEM)研究了HP-β-CD与虎杖甙的主客体作用,并考察了包合作用对虎杖甙光稳定性的影响。结果表明,HP-β-CD与虎杖甙形成了包合比为1:1的包合物,缔合常数为1308 mol-1·L;包合后虎杖甙在水中的溶解度提高10倍以上,热稳定性提高,分解温度从270 ℃提高至298 ℃;光稳定性实验表明包合后虎杖甙的光稳定性得到了有效提高。这为虎杖甙在食品、化妆品等领域的使用提供了新的方法和信息。

Abstract:

The inclusion complex between piceid and (2-hydroxy)propyl-β-cyclodextrin (HP-β-CD) was prepared by freeze-drying method. The effects of the inclusion behavior of piceid with HP-β-CD was investigated by methods of phase-solubility, X-ray diffraction (XRD), thermogravimetric analysis (TG), differential scanning calorimetry (DSC), and scanning electron microscopy (SEM). Meanwhile, the photostability of piceid and its inclusion complex was also investigated. The results showed that piceid formed 1:1 stoichiometric inclusion complex with HP-β-CD, and the apparent stability constant was 1308 mol-1·L. The solubility of the piceid was enhanced more than 10 times. Also the decomposition temperature of piceid was improved from 270 ℃ to 298 ℃ after encapsulated by HP-β-CD. Furthermore, the photostability of the inclusion complex was more stable than the native piceid. Therefore, it can be a potentially promising way to promote its food supplement or cosmetic preparations.
